General description
KARS, also known as Lysyl-tRNA synthetase (LysRS), is the enzyme that connects the lysine to its specific tRNA for use in protein translation. KARS is also a powerful inflammatory agent when released into extracellular matrix as one might expect. KARS is largely cytoplasmically localized or localized to internal membranes but isoforms are found in the mitochondria and nucleus as well. KARS became the focus of much attention with HIV-1 as KARS interacted with HIV-1's GAG protein and facilitated the selective packaging of tRNA3(lys) nucleic acid which acted as the primer for viral reverse transcription and initiation of viral gene expression. There is also at least one genetic disease linked to KARS. It is the cause of Charcot-Marie-Tooth neuropathy a progressive peripheral nervous system disease characterized by progressive weakness and atrophy of the muscles and demyelination and deterioration the peripheral nerves and axons. EMD-Millipore's Anti-KARS mouse monoclonal antibody has been tested in western blot on recombinant protein and in paraffin embedded immunohistochemistry on human cervical carcinoma tissue sections.
Immunogen
Purified recombinant fragment of human KARS expressed in E. Coli.
Physical form
Mouse monoclonal IgG1 ascitic fluid containing up to 0.1% sodium azide.
Unpurified
Quality
Evaluated by Western Blotting in truncated Trx-KARS recombinant protein, truncated MBP-KARS and full length KARS transfected CHO-K1 cell lysates.
Western Blotting Analysis: A 1:500-2,000 dilution of this antibody detected KARS in truncated Trx-KARS recombinant protein, truncated MBP-KARS and full length KARS transfected CHO-K1 cell lysates.
Storage and Stability
Stable for 1 year at -20°C from date of receipt.
Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
Target description
~24, ~44, and ~50 kDa observed. This antibody recognizes truncated MBP-KARS at ~24 kDa, full length KARS at ~44 kDa, and transfected CHO-K1 cell lysate at ~50 kDa
