General description
Polymer-based particles for reversed-phase (RP) HPLC generally have two benefits over silica-based particles: They are more tolerant of high and low pH operating conditions and they do not have the silanol interactions that cause peak tailing of some basic analytes. Also, some silicas have an intrinsic metal content that may cause chelation problems with sensitive biomolecules, leading to poor recovery. Polymeric TSKgel columns come in C18 and phenyl functional groups and a range of particle porosities. Fast kinetics and quantitative protein recovery at submicrogram loading are characteristic of nonporous resin-based TSKgel Octadecyl-NPR columns. Highly cross-linked polymethacrylate base material of TSKgel Octadecyl-4PW provides excellent stability in high pH buffer systems and can withstand rigorous cleaning with either acid or base. The 500Å pore allows for anaylsis of proteins up to 200kDa while the particle size offerings allow for analytical, semi-preparative and preparative scale separations.
