Analysis Note

ControlMouse pancreas tissue lysate

Application

Use Anti-LRH-1 Antibody, clone 1LRH-2E1 (Mouse Monoclonal Antibody) validated in WB, ICC to detect LRH-1 also known as Liver receptor homolog 1.

Western Blot Analysis: A 1:1,000 dilution from representative lot detected Liver receptor homolog-1 in HepG2 and HeLa cell lysates (M. Oulad, IGBMC, France.).

Immunocytochemistry Analysis: A representative lot detected Liver receptor homolog-1 in HepG2 and HeLa cells (M. Oulad, IGBMC, France.).

General description

Liver receptor homolog 1 (LRH-1, CPF, FTF, PHR-1, FTF, or hB1F) is an orphan nuclear receptor that is highly expressed in the intestines, pancreas, ovary, and liver. LRH-1 is a constitutively active transcription factor with a complex role in many cellular processes which may include lipid metabolism, cell proliferation, development, reproduction, and immunity. LRH-1 may interact with an array of transcriptional coactivators that modulate its activity, among which are the MBF1, SRC, and PGC-1α proteins. In addition, ERK phosphorylation on Ser-238 and Ser-243 also enhances LRH-1 activity. However, LRH-1 is also negatively regulated by several factors. For instance, corepressors such as SHP, DAX-1, and PROX-1, preclude its binding to DNA; and sumoylation promotes its inactivation and sequestration into nuclear bodies. LRH-1 targets many genes including the CYP7A1 and CYP8B1 genes involved in bile synthesis; and Oct4 required for pluripotency in embryonic stem cells. LRH-1 has been implicated in inflammatory bowel disease, and tumor development in the intestines and pancreas.

Immunogen

Linear peptide corresponding to the mouse Liver receptor homolog-1.

Physical form

Mouse monoclonal IgG2aλ ascites with 0.05% sodium azide.

Quality

Evaluated by Western Blot in mouse pancreas tissue lysate.

Western Blot Analysis: A 1:1,000 dilution of this antibody detected Liver receptor homolog-1 in 10 µg of mouse pancrease tissue lysate.

Target description

~64 kDa, ~56 kDa, and ~42 kDa observed. This antibody detects three isoforms of Liver receptor homolog-1 as described by Uniprot No. O00482.
