Biochem/physiol Actions

Histone deacetylase 2 (HDAC2) is known to regulate DNA damage signalling, maintain genomic stability and prevents tumor formation and progression. Hdac2 does not possess a DNA binding domain, it forms multi-component repressor complexes for its interaction with transcription factors. Hdac2 also mediates cell proliferation, differentiation and apoptosis. HDAC2 controls response to chronic inflammation in COPD (chronic obstructive pulmonary disease). It activates Wnt signaling pathway via β-catenin T-cell transcription factor. Deletion of HDAC2 along with HDAC1 leads to anemia and thrombocytopenia.

General description

Human HDAC2, GenBank Accession No. Q92769, full length with C-terminal DDDDK tag (FLAG), MW = 60 kDa, expressed in baculovirus expression system. The HDAC2 (histone deacetylase 2) gene is mapped to human chromosome 6q21 and is ubiquitously expressed.

Physical form

Formulated in 25 mM Tris-HCl, pH 8.0, 138 mM NaCl, 0.05% Tween-20 and 10% glycerol.

Preparation Note

Thaw on ice. Upon first thaw, briefly spin tube containing enzyme to recover full content of the tube. Aliquot enzyme into single use aliquots. Store remaining undiluted enzyme in aliquots at -70°C. Note: Enzyme is very sensitive to freeze/thaw cycles.

Unit Definition

One unit is defined as the amount of enzyme required to deacetylate 1 pmol of substrate/min at 37°C.
