Sestrin 1 (SESN1) activates AMP-responsive protein kinase (AMPK) which phosphorylates tuberous sclerosis complex 2 (TSC2), and subsequently inhibits mammalian target of rapamycin protein (mTOR). Mutations in the gene encoding the protein have been linked to heterotaxia.

Sestrin 1 (SESN1) is expressed in the nucleus and is a target gene of the tumor suppressor p53. The gene encoding it is localized on human chromosome 6q21.
