Mitogen-activated protein kinase 1 (MEK1) participates in MAPK/ ERK (extracellular signal-regulated kinases) pathway, which plays a vital role in regulation of cell proliferation, differentiation and survival. Mutation of MEK1 gene leads to non-small cell lung cancer (NSCLC). In addition, aberrations of the gene is also observed in adenocarcinoma patients with current or former smoking status. MEK1 influences ERK activation in MAPK pathway and induce proliferation and anti-apoptotic signals. MEK1 facilitates platinum resistance via activation of excision repair cross-complementation group 1 (ERCC1) protein. Clinical trials show that, Trametinib acts as a MEK1/MEK2 inhibitor and is used to treat various cancers caused by MEK1 mutation including melanoma, colorectal cancer.

General description

Mitogen-activated protein kinases 1 (MEK1) is encoded by the gene mapped to human chromosome 15q22.31 and the gene contains 11 exons with intervening sequences. MEK1 belongs to the MEK protein family and members of this family contain kinase domain at their amino-terminal and a catalytic domain at their carboxyl-terminal end.
